凈化(hua)工(gong)作(zuo)臺是一種局(ju)部(bu)層(ceng)流裝置,能在局(ju)部(bu)形成高(gao)潔(jie)度的工(gong)作(zuo)環境。它(ta)由(you)工(gong)作(zuo)臺、過(guo)濾(lv)器、風機、靜壓(ya)箱和支撐體(ti)等組(zu)成,采(cai)用(yong)過(guo)濾(lv)空(kong)氣使(shi)工(gong)作(zuo)臺操作(zuo)區達到凈化(hua)除(chu)菌(jun)的目的。室內空(kong)氣經預過(guo)濾(lv)器和高(gao)效過(guo)濾(lv)除(chu)塵后以垂直或水平(ping)層(ceng)流狀態通過(guo)工(gong)作(zuo)臺的操作(zuo)區,由(you)于空(kong)氣沒(mei)有(you)渦流,所以,任何一點灰塵或附著在灰塵上的雜(za)菌(jun)都能被(bei)排除(chu),不易向別處擴散和轉移。因(yin)此,可使(shi)操作(zuo)區保持無菌(jun)狀態。
與沒有細菌室和疫苗注射箱較好,采用進行過濾清潔辦公臺具備有辦公能力好、控制便宜、沒有細菌郊果安全、無進行維護清潔藥物對人體內的影響、占有面積計算小且可中移動等優越性。這樣放著沒有細菌內采用,沒有細菌郊果最好。其缺欠是產品報價比較貴,預進行過濾清潔芯和效率進行過濾清潔芯還可以死期進行維護清潔和修改。 各類高壓蒸汽加熱消毒鍋高壓(ya)蒸(zheng)(zheng)汽滅(mie)菌鍋是一(yi)個(ge)密閉的(de)(de)、可(ke)以(yi)耐受一(yi)定(ding)壓(ya)力(li)的(de)(de)雙層金屬鍋。鍋底或夾(jia)層內盛水,當水在(zai)鍋內沸(fei)騰時(shi)由于蒸(zheng)(zheng)汽不能逸出,使鍋內壓(ya)力(li)逐漸升(sheng)高,水的(de)(de)沸(fei)點(dian)和溫度可(ke)隨之(zhi)升(sheng)高,從而達到高溫滅(mie)菌的(de)(de)目的(de)(de)。一(yi)般(ban)在(zai)0.11MPa的(de)(de)壓(ya)力(li)下,121℃滅(mie)菌20~30min,包括芽孢在(zai)內的(de)(de)所有微生物均可(ke)被殺死(si)。如果(guo)滅(mie)菌物品(pin)體積較大,蒸(zheng)(zheng)汽穿透(tou)困難,可(ke)以(yi)適當提高蒸(zheng)(zheng)汽壓(ya)力(li)或延長滅(mie)菌時(shi)間。
各類髙壓髙壓蒸汽式消毒鍋有水平、開式、臺式式等多重類行,在生物學體學實驗所室,最最經常使用的是臺式式和開式各類髙壓蒸汽式髙壓蒸汽式消毒鍋。和過熱蒸汽髙壓蒸汽式消毒鍋比起,各類髙壓髙壓蒸汽式消毒鍋的特征是髙壓蒸汽式消毒需用的時短、節約用電然料、髙壓蒸汽式消毒完全等。其問題是房價昂貴,髙壓蒸汽式消毒存儲容量較小。 教育培養箱培(pei)(pei)養(yang)(yang)箱(xiang)是(shi)培(pei)(pei)養(yang)(yang)微生物(wu)(wu)的(de)(de)專用(yong)設備。制熱(re)式培(pei)(pei)養(yang)(yang)箱(xiang)是(shi)由電爐絲和(he)溫(wen)度(du)控(kong)制儀合成的(de)(de)固定(ding)體積(ji)的(de)(de)恒(heng)溫(wen)培(pei)(pei)養(yang)(yang)裝置(zhi),大小規格不一。微生物(wu)(wu)實驗室(shi)常用(yong)的(de)(de)培(pei)(pei)養(yang)(yang)箱(xiang)工作容(rong)積(ji)有450×450×350mm3或650×500×500mm3,適用(yong)于室(shi)溫(wen)至(zhi)60℃之間的(de)(de)各(ge)類微生物(wu)(wu)培(pei)(pei)養(yang)(yang)。目(mu)前,隨著科學(xue)水平的(de)(de)發展,培(pei)(pei)養(yang)(yang)箱(xiang)設備的(de)(de)完善程度(du)和(he)價(jia)格有很大差別。有各(ge)種結構合理、功(gong)能齊全的(de)(de)培(pei)(pei)養(yang)(yang)箱(xiang),如(ru)恒(heng)溫(wen)培(pei)(pei)養(yang)(yang)箱(xiang)、恒(heng)溫(wen)恒(heng)濕培(pei)(pei)養(yang)(yang)箱(xiang)、低(di)溫(wen)培(pei)(pei)養(yang)(yang)箱(xiang)、微生物(wu)(wu)多用(yong)培(pei)(pei)養(yang)(yang)箱(xiang)和(he)二氧化碳培(pei)(pei)養(yang)(yang)箱(xiang)等。有的(de)(de)用(yong)計(ji)算機(ji)控(kong)制,可選擇(ze)多條時間線變換溫(wen)差,從而克服了環境溫(wen)度(du)的(de)(de)影(ying)響,一年四季均能達到培(pei)(pei)養(yang)(yang)要求的(de)(de)溫(wen)度(du)。
微(wei)生物多用培養箱是集(ji)加熱、制冷和振蕩(dang)于一體(ti)的(de)微(wei)生物液(ye)體(ti)發(fa)酵裝(zhuang)置。工(gong)作(zuo)室的(de)溫度在(zai)(zai)15~50℃范圍內(nei)任意(yi)選(xuan)定,選(xuan)定后經溫控儀自動控制,保(bao)持工(gong)作(zuo)室內(nei)恒溫。同時設有可控硅調(diao)速系統(tong),振蕩(dang)機轉(zhuan)速可在(zai)(zai)1~220rpm范圍內(nei)任意(yi)調(diao)控。
常溫、烘干箱干(gan)燥箱是用(yong)(yong)于除(chu)去潮(chao)濕物(wu)料內及器(qi)(qi)皿內外水分或(huo)(huo)其它(ta)揮發性(xing)溶(rong)液的(de)(de)設備。類型很多,有(you)(you)(you)箱式(shi)(shi)、滾筒式(shi)(shi)、套間(jian)式(shi)(shi)、回轉(zhuan)式(shi)(shi)等。微(wei)生物(wu)學實驗室多用(yong)(yong)箱式(shi)(shi)干(gan)燥箱,大小規(gui)格不一。工作室內配(pei)有(you)(you)(you)可(ke)活動的(de)(de)鐵絲網(wang)板(ban),便于放置被干(gan)燥的(de)(de)物(wu)品(pin)。制(zhi)(zhi)熱升(sheng)溫式(shi)(shi)干(gan)燥箱也是有(you)(you)(you)電爐絲和(he)溫度(du)控(kong)(kong)制(zhi)(zhi)儀組成(cheng),可(ke)調節溫度(du)從室溫至300℃任意(yi)選擇(ze)。有(you)(you)(you)的(de)(de)干(gan)燥箱采(cai)用(yong)(yong)導電溫度(du)計(ji)為(wei)敏感(gan)元件,配(pei)合晶體管(guan)(guan)和(he)繼(ji)電器(qi)(qi)組成(cheng)自動控(kong)(kong)制(zhi)(zhi)系統,克(ke)服了金屬管(guan)(guan)型熱膨脹控(kong)(kong)制(zhi)(zhi)的(de)(de)缺點。此(ci)外,還(huan)有(you)(you)(you)真空(kong)干(gan)燥箱(配(pei)有(you)(you)(you)真空(kong)泵和(he)氣壓表),可(ke)在常(chang)壓或(huo)(huo)減壓下操作。
搖床
搖(yao)(yao)床(chuang)(chuang)又稱(cheng)搖(yao)(yao)瓶(ping)機,它是培(pei)養好氣性微(wei)生(sheng)物的(de)(de)小型試(shi)驗設備(bei)或作為(wei)種(zhong)(zhong)子(zi)擴大培(pei)養之(zhi)(zhi)用,常用的(de)(de)搖(yao)(yao)床(chuang)(chuang)有(you)往(wang)復式和旋轉(zhuan)式兩種(zhong)(zhong)。往(wang)復式搖(yao)(yao)床(chuang)(chuang)的(de)(de)往(wang)復頻率一般在(zai)80~140次/min,沖(chong)程一般為(wei)5~14cm,如(ru)頻率過快、沖(chong)程過大或瓶(ping)內液體(ti)裝(zhuang)量(liang)過多(duo),在(zai)搖(yao)(yao)動(dong)時(shi)液體(ti)會濺到包扎(zha)瓶(ping)口的(de)(de)紗布(bu)或棉塞上,導致雜菌污(wu)染,特別是啟動(dong)時(shi)更容(rong)易發生(sheng)這(zhe)種(zhong)(zhong)情況。旋轉(zhuan)式搖(yao)(yao)床(chuang)(chuang)的(de)(de)偏心距(ju)一般在(zai)3~6cm之(zhi)(zhi)間,旋轉(zhuan)次數為(wei)60~300rpm。
放在(zai)搖床上的(de)(de)培養瓶(ping)(ping)(一(yi)般為(wei)三(san)角瓶(ping)(ping))中(zhong)的(de)(de)發酵液所需要的(de)(de)氧(yang)是由空氣經瓶(ping)(ping)口包扎的(de)(de)紗(sha)布(一(yi)般8層)或棉塞通入的(de)(de),所以氧(yang)的(de)(de)傳遞與瓶(ping)(ping)口的(de)(de)大小、瓶(ping)(ping)口的(de)(de)幾(ji)何形狀、棉塞或紗(sha)布的(de)(de)厚度和密(mi)度有關。在(zai)通常(chang)情況下,搖瓶(ping)(ping)的(de)(de)氧(yang)吸(xi)收系數取決于搖床的(de)(de)特性(xing)和三(san)角瓶(ping)(ping)的(de)(de)裝樣量。

疫苗接種箱接(jie)種(zhong)(zhong)箱(xiang)分(fen)為(wei)固體菌種(zhong)(zhong)接(jie)種(zhong)(zhong)箱(xiang)和液體菌種(zhong)(zhong)接(jie)種(zhong)(zhong)箱(xiang)兩(liang)種(zhong)(zhong)。固體菌種(zhong)(zhong)接(jie)種(zhong)(zhong)箱(xiang)是一(yi)個用木(mu)料和玻(bo)(bo)(bo)璃(li)制成或(huo)由有機玻(bo)(bo)(bo)璃(li)焊接(jie)而成的(de)密閉小(xiao)箱(xiang)。又分(fen)為(wei)雙人和單人操(cao)作(zuo)箱(xiang)。箱(xiang)體可(ke)大可(ke)小(xiao),一(yi)般箱(xiang)體長約143cm,寬86cm,總高(gao)154cm,支架(jia)76cm。箱(xiang)的(de)上部(bu)左右兩(liang)側各(ge)裝有兩(liang)扇(shan)能啟閉的(de)玻(bo)(bo)(bo)璃(li)推拉門,方便菌種(zhong)(zhong)進(jin)出。窗的(de)下部(bu)分(fen)別設有兩(liang)個直(zhi)徑(jing)約13cm的(de)圓洞,兩(liang)洞的(de)中心距離為(wei)52cm(同肩寬),洞口裝有帶松緊帶的(de)袖(xiu)套(tao),以防雙手(shou)(shou)在箱(xiang)內操(cao)作(zuo)時(shi),外界(jie)空氣進(jin)入箱(xiang)內造(zao)成污染。操(cao)作(zuo)時(shi)兩(liang)人相對而坐,雙手(shou)(shou)通(tong)過(guo)袖(xiu)套(tao)伸入箱(xiang)內。箱(xiang)兩(liang)側最好(hao)也裝上玻(bo)(bo)(bo)璃(li),箱(xiang)頂部(bu)為(wei)木(mu)板或(huo)玻(bo)(bo)(bo)璃(li)。箱(xiang)內頂部(bu)裝有紫外線殺菌燈和照明用日光燈各(ge)一(yi)支。箱(xiang)體安裝木(mu)板或(huo)玻(bo)(bo)(bo)璃(li)均可(ke),但(dan)要注意密封(feng)。
液體(ti)菌(jun)種(zhong)接種(zhong)箱(xiang)是專為(wei)移接液體(ti)菌(jun)種(zhong)而設計的(de)。比固體(ti)菌(jun)種(zhong)箱(xiang)窄長,單側兩人操作。內設軌(gui)道和紫(zi)外線燈,箱(xiang)兩端開有高25cm,寬10cm的(de)長方(fang)(fang)形出(chu)(chu)(chu)口,方(fang)(fang)便菌(jun)種(zhong)進出(chu)(chu)(chu),洞口設有小推(tui)門。進出(chu)(chu)(chu)口下處(chu)設蒸汽源,接種(zhong)時用蒸汽封(feng)住進出(chu)(chu)(chu)口,以(yi)防(fang)雜菌(jun)進入箱(xiang)內。箱(xiang)背面設有液體(ti)菌(jun)種(zhong)移接管能進入的(de)小孔。
接(jie)(jie)種箱(xiang)滅菌(jun)時(shi),用(yong)紫(zi)外(wai)(wai)線(xian)照射30min。如果沒有紫(zi)外(wai)(wai)線(xian)燈,可用(yong)甲(jia)醛和(he)高(gao)錳(meng)酸鉀(甲(jia)醛10~14mL/m3+高(gao)錳(meng)酸鉀5~7g/m3空(kong)間)熏蒸30min以上。使用(yong)時(shi),先將所需物(wu)品和(he)工(gong)具放(fang)入接(jie)(jie)種箱(xiang)內(nei),然后進(jin)行(xing)藥劑熏蒸和(he)紫(zi)外(wai)(wai)線(xian)滅菌(jun),再按無菌(jun)操(cao)(cao)作(zuo)進(jin)行(xing)接(jie)(jie)種。接(jie)(jie)種箱(xiang)的結構簡單,造價低廉(lian),易消毒(du)滅菌(jun),操(cao)(cao)作(zuo)方便,而且人在箱(xiang)外(wai)(wai)操(cao)(cao)作(zuo),氣溫較高(gao)時(shi)也能作(zuo)業。缺點是進(jin)出培養基(ji)費工(gong)費時(shi),每(mei)次接(jie)(jie)種前(qian)都需要進(jin)行(xing)滅菌(jun)。
墻上 微生物實驗室的冰箱主要有兩種:普通冰箱和低溫冷凍冰箱。普通冰箱一般都具有兩個柜子,即鮮藏柜和冷藏柜,溫度分別為4℃和-20℃;低溫冷凍冰箱溫度一般控制在-40~-80℃。它們都可以用于微生物菌種保藏。鮮藏柜常用于保存斜面菌種,保藏時間在3個月左右。超過3個月,斜面就會變干,因此需要轉接菌種。如果要長時間保存菌種,則需要經過處理后,貯藏于普通冰箱的冷藏柜或低溫冷凍冰箱中,它們的保藏時間較長,一般都在1年以上。
